Combine broccoli and water in a microwave-safe shallow dish. Cover with vented plastic wrap and microwave on High for 2 to 3 minutes or just until tender. (Or in a covered small saucepan, cook broccoli in small amount of boiling water for 4 to 6 minutes or just until tender.) Drain well; set aside.
Layer half of the bread cubes in a greased 2-quart square baking dish. Top with cheese, ham and broccoli; add remaining bread cubes. Combine milk, egg, onion, mustard and pepper in a medium bowl. Pour egg mixture evenly over the layers in dish. Cover and refrigerate for 2 to 24 hours.
Preheat oven to 325°F. Uncover dish and bake for 60 to 70 minutes or until knife inserted near center comes out clean. Let stand for 5 minutes.
